PRINCIPLE
In acid medium, acid phosphatase hydrolyzes the substrate producing α-naphtol that reacts with the diazonium salt forming an azo dye and changing the Abs of the reagent.In optimum conditions, the ΔAbs/min is directly related to the concentration of acid phosphatase of the sample.
DIAGNOSTIC USE
Acid phosphatase, especially the tartrate labile fraction or prostatic phosphatase (PAP), is elevated in cases of metastatic prostate cancer. The concentration does not increase in the absence of metastasis or it slightly does.High values are also found in multiple myeloma, in some lymphoblastic leukemias and osteolytic bone metastases.Single test result can not be used to make a clinical diagnosis. It should integrate clinical and laboratory data.
